ABSTRACT

Objective? To?systematically?analyze?the?effect?of?haemoperfusion?(HP)?combined?with?continuous?veno-veno?haemofiltration?(CVVH)?in?the?treatment?of?the?patients?with?paraquat?poisoning?(PQP).? Methods? Words?of?paraquat,?poisoning,?continuous?venous?hemofiltration,?hemoperfusion,?hemodiafiltration?in?Chinese?and?paraquat,?poisoning,?intoxication,?haemofiltration,?continuous?venovenous?haemofiltration,?haemoperfusion?in?English?were?chosen?as?keywords,?the?Chinese?and?English?literatures?about?acute?PQP?treated?with?HP?combined?with?CVVH?published?in?Wanfang?database,?CNKI,?CBM,?VIP?database,?PubMed,?Embase,?Cochrane?Library?were?searched?by?computer,?and?the?retrieval?time?was?from?the?establishment?of?the?database?to?July?2018.?The?experimental?group?was?treated?with?HP?combined?with?CVVH,?while?the?control?group?was?treated?with?HP?alone.?Besides,?the?outcome?indicators?included?mortality,?survival?time?of?dead?patients?(the?patient's?time?from?exposure?to?poison?to?death),?serum?creatinine?(SCr),?alanine?aminotransferase?(ALT),?arterial?partial?pressure?of?oxygen?(PaO2),?and?incidence?of?circulatory?and?respiratory?failure.?The?literature?data?were?extracted?by?two?researchers?independently,?the?quality?of?the?literature?was?evaluated? according?to?the?modified?Jadad?score?table?or?Newcastle-Ottawa?scale?(NOS),?and?the?Meta-analysis?was?carried?out?by?RevMan?5.3?software;?and?the?stability?of?the?results?of?Meta-analysis?was?tested?by?sensitivity?analysis.?Further,?the?publication?bias?was?analyzed?through?drawing?a?funnel?diagram.? Results? Finally,?20?articles?were?included,?with?18?in??Chinese?and?2?in?English.?Among?them,?6?were?randomized?controlled?trial?(RCT)?and?14?were?case-control?studies.?Furthermore,?a?total?of?2?870?patients?were?involved,?with?1?558?in?the?control?group?and?1?312?in?the?experimental?group.?Meta-analysis?showed?that?the?mortality?rate?of?patients?in?the?experimental?group?was?significantly?lower?than?that?in?the?control?group?[odds?ratio?(OR)?=?0.55,?95%?confidence?interval?(95%CI)?=?0.42?to?0.73,?P <?0.000?1],?the?patients'?time?from?toxin?exposure?to?death?was?significantly?longer?than?that?in?the?control?group?[standard?mean?difference?(SMD)?=?2.16,?95%CI?=?1.46?to?2.86,?P?<?0.000?01).?In?the?course?of?treatment,?the?peak?value?of?SCr?in?the?experimental?group?was?significantly?lower?than?that?in?the?control?group?(SMD?=?-0.53,?95%CI?=?-0.65?to?-0.42,?P?<?0.000?01),?and?the?peak?value?of?ALT?was?also?decreased?(SMD =?-0.72,?95%CI?=?-0.99?to?-0.44,?P?<?0.000?01).?Besides,?there?was?no?significant?difference?in?PaO2?between?the?two?groups?on?the?3rd?day?of?treatment?(SMD =?0.15,?95%CI?=?-0.19-0.49,??P?=?0.40),?but?on?the?7th?day,?PaO2?in?the?experimental?group?was?significantly?higher?than?that?in?the?control?group?(SMD?=??0.23,?95%CI =?0.29?to?0.98,?P?=?0.000?3).?Furthermore,?the?incidence?of?circulatory?failure?in?the?experimental?group?was?significantly?lower?than?that?in?the?control?group?(OR =?0.26,?95%CI?=?0.19?to?0.37,?P?<?0.000?01),?but?the?incidence?of?respiratory?failure?was?significantly?higher?than?that?in?the?control?group?(OR =?4.14,?95%CI?=?3.00?to?5.72,?P?<?0.000?01).?The?influence?of?heterogeneity?on?statistical?results?was?excluded?in?the?sensitivity?analysis,?and?funnel?plot?diagram?was?applied?to?indicate?the?publication?bias?of?mortality?and?survival?time?of?the?dead?patients.? Conclusion? Combined?with?HP?alone,?HP?combined?with?CVVH?could?better?improve?liver?and?kidney?function?and?oxygenation?state?of?PQP?patients,??reduce?the?incidence?of?early?circulatory?failure,?prolong?the?survival?time?and?reduce?the?death?rate?of?PQP?patients.

ABSTRACT

Objective To compare the influence of sevoflurane inhalation sedation and propofol intravenous sedation on duration of endotracheal intubation as well as the length of intensive care unit (ICU) stay and total length of hospital stay in postoperative critical patients.Methods Six databases including CNKI,Wanfang data,PubMed,Embase,Cochrane Library and Web of Science were searched for randomized controlled trials (RCTs) about the influence of sevoflurane inhalation sedation or propofol intravenous sedation on the sedation time,the duration of endotracheal intubation,the length of ICU stay,the total length of hospital stay and the adverse effects rate in postoperative critical patients from the time of database establishment to July 2018.At the same time,the reference materials of included literature were retrieved manually.All literatures were screened by three independent reviewers,and the data extraction and quality evaluation of the included studies were conducted.Meta-analysis was used for RCT that met the quality standards.Results A total of 7 RCT studies were enrolled involving 537 patients who were all transferred into ICU after surgery with trachea cannula.Among the patients,272 received sevoflurane sedation while the other 265 received propofol sedation.All the included studies were well designed and of high quality.The results of Meta-analysis showed that compared with propofol sedation,sevoflurane sedation could significantly shorten the duration of endotracheal intubation [standardized mean difference (SMD) =-0.60,95% confidence interval (95%CI) =-0.88 to-0.31,P < 0.000 1]and the total length of hospital stay (SMD =-0.36,95%CI =-0.61 to-0.12,P =0.003),and lower the cardiac troponin T (cTnT) within 12-24 hours after ICU admission (SMD =-0.61,95%CI =-0.85 to-0.36,P < 0.000 01).There was no significant difference in the sedation time (SMD =-0.07,95%CI =-0.29 to 0.15,P =0.52),the length of ICU stay (SMD =-0.19,95%CI =-0.39 to 0.01,P =0.06),the incidence of nausea and vomiting [odds ratio (OR) =1.19,95%CI =0.61 to 2.32,P =0.61] or incidence of delirium (OR =0.80,95%CI =0.34 to 1.90,P =0.62) between sevoflurane group and propofol group.Conclusions Sevoflurane inhalation sedation may lead to shorter duration of endotracheal intubation and total length of hospital stay,and had better protection for myocardium as compared with propofol intravenous sedation.The above conclusions needed further study to confirm,due to the lack of literature enrolled in this Meta-analysis.

Exploratory experiment is the key to the experimental physiology,which is universally applicable training of innovative talents.More importantly,it is an important way to cultivate innovative talents of medicine.On the basis of the original multi-subject evaluation system,we have formed a comprehensive evaluation system through refining the scoring item for teachers and students,enriching the forms of student evaluation,and building the student self-assessment.This comprehensive evaluation system is set to promote the development of experimental physiology,especially the exploratory experiments.The practice of this evaluation system effectively improve the objectivity of the evaluation,provide a basis for reform of exploratory experiment,and then promote the training of innovative talents.

ABSTRACT

AIM:To investigate the effect of ionizing radiation on epithelial-mesenchymal transition in lung cancer cell line A549 and its possible mechanism.METHODS:The lung cancer A549 cells were irradiated with different doses (0 Gy, 1 Gy, 2 Gy, 4 Gy and 8 Gy) of X-ray for different time.The morphological changes of the cells were observed under inverted microscope at time points of 12 h, 24 h and 48 h.The expression of vimentin, N-cadherin, E-cadherin and transcription factor c-Myc was detected by Western blot at the time points of 12 h and 24 h.RESULTS:After ionizing radiation, the contours of the A549 cells were unclear, the protrusions increased, and the edges were irregular, with fried egg-like collapses.The mesenchymal morphology of the A549 cells was most obvious after irradiation at 8 Gy for 48 h.Compared with 0 Gy irradiation group, the expression of vimentin was down-regulated seemingly 12 h after irradiation, but up-regulated in 2 Gy, 4 Gy and 8 Gy irradiation groups for 24 h, and the most obvious effect was observed in 2 Gy irradiation group (P<0.01).Compared with 0 Gy irradiation group, the expression of N-cadherin was up-regulated in 1 Gy, 2 Gy and 4 Gy irradiation groups for 24 h (P<0.05), while the expression of E-cadherin was not influenced.The up-regulation of vimentin expression in lung cancer cell line A549 was positively correlated with c-Myc expression.CONCLUSION:Ionizing radiation may promotes epithelial-mesenchymal transition in the lung cancer cell line A549 by up-regulating the c-Myc expression.

[ ABSTRACT] AIM:To compare the differences of the genome-wide methylation levels and methylated regions be-tween nasopharyngeal carcinoma ( NPC) cells in the same genetic background but different radiation resistance ( CNE-2 cells and CNE-2R cells).METHODS:Using the method which was developed by Doctor Zhao Cun-you, based on using methyl-sensitive restriction enzyme to measure the genome-wide methylation levels.In addition, MeDIP-Seq was used to analyze the methylated regions in 6 gene functional elements, including the upstream 2k sequence, 5’ UTR, coding se-quence, intron, 3’UTR and downstream 2k sequence, between CNE-2 cells and CNE-2R cells.RESULTS:The genome-wide methylation level was approximately 30%lower in CNE-2R cells than that in CNE-2 cells.No obvious difference on the amount of genes and the coverage of the peak in the 6 gene functional elements was observed.However, the methylation pattern of plentiful genes had altered in the gene function elements.CONCLUSION:The genome-wide methylation levels and methylated regions between NPC cells in the same genetic background but different radiation resistance were quite dif-ferent, indicating that the DNA methylation may be associated with NPC radioresistance.

AIM:To investigate the expression of C/EBP homologous protein (CHOP) and its correlation with proliferative/apoptotic ratio (PAR) in colorectal adenoma-carcinoma sequence under the same genetic background .ME-THODS:Four kinds of tissue samples under the same genetic background from 23 patients, including normal colorectal tissue, adenoma with low-grade intraepithelial neoplasia , adenoma with high-grade intraepithelial neoplasia and colorectal adenocarcinoma samples , were collected .TUNEL method and Ki-67 immunohistochemistry were applied to determine the PAR.The expression of CHOP was detected by immunohistochemistry SABC method .RESULTS: ( 1 ) Under the same genetic background , the level of CHOP expression is significantly higher in colorectal adenocarcinoma than that in the ade -noma with high-grade intraepithelial neoplasia , the adenoma with low-grade intraepithelial neoplasia and the normal muco-sa.The level of CHOP expression was significantly higher in the adenoma with high -grade intraepithelial neoplasia than that in the adenoma with low-grade intraepithelial neoplasia and the normal mucosa .The level of CHOP expression was signifi-cantly higher in the adenoma with low-grade intraepithelial neoplasia than that in normal mucosa .(2) Under the same ge-netic background , PAR was significantly higher in the colorectal adenocarcinoma than that in the adenoma with high -grade intraepithelial neoplasia , the adenoma with low-grade intraepithelial neoplasia and the normal mucosa .PAR was significant-ly higher in the adenoma with high-grade intraepithelial neoplasia than that in the adenoma with low-grade intraepithelial neoplasia and the normal mucosa .PAR was significantly higher in the adenoma with low-grade intraepithelial neoplasia than that in the normal mucosa.(3) CHOP levels were positively correlated with PAR in the adenoma with low-grade intraepi-thelial neoplasia , adenoma with high-grade intraepithelial neoplasia and colorectal adenocarcinoma .CONCLUSION:CHOP expression and PAR continuously increased and positively correlated along the adenoma -carcinoma sequence , indica-ting that endoplasmic reticulum stress mediates the carcinogenesis of colorectal adenomas .

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the inhibitory effect of deuterium-depleted water (DDW) on the proliferation of nasopharyngeal carcinoma (NPC) cells in vitro and explore the possible mechanism.</p><p><b>METHODS</b>The growth inhibition of NPC cells and preosteoblast MC3T3-E1 cells following DDW treatment was measured by MTT assay and plate colony formation assay. The changes in migration and invasion of NPC cells were evaluated using Transwell and boyden chamber assays. The protein expression of proliferating cell nuclear antigen (PCNA) was determined using Western blotting. Flow cytometry was employed to evaluate the changes in cell cycle distribution after DDW treatment.</p><p><b>RESULTS</b>DDW with deuterium concentrations of 100, 75 and 50 ppm significantly suppressed the cell proliferation (P<0.05) and lowered colony formation capacity and invasiveness of the NPC cells (P<0.01). Western blotting demonstrated a down-regulated expression of PCNA in the cells by DDW. DDW also caused obvious cell cycle arrest in the NPC cells with reduced cells in S phase and significantly increased cells in G(1) phase (P<0.05). Rather than causing growth inhibition, DDW promoted the growth of normal control MC3T3-E1 cells.</p><p><b>CONCLUSION</b>DDW possesses selective biological effects to inhibit the proliferation of NPC cells in vitro, suggesting the potential of DDW as a novel nontoxic adjuvant therapeutic agent in antitumor therapy.</p>

ABSTRACT

Deuterium is an important predisposing factor for cancer. Deuterium-depleted Water, also known as low deuterium water, ultra-light water or no deuterium water, can be obtained by removing deuterium from natural water. Studies have shown that water with a low deuterium concentration (<65% percent of volume) can inhibit cancer growth. Clinical trials demonstrated that drinking DDW (10-20 ppm) caused growth arrest of malignant cells in cancer patients and significantly prolonged the patient survival with also improved quality of life. A wide range of anti-cancer drugs in current use are associated with severe adverse effects, while deuterium-depleted water appears to have virtually no pharmacological side effects and is convenient to administer. The authors review the advances in the researches of anti-cancer effects and the underlying mechanisms of deuterium-depleted water.

ABSTRACT

AIM:To investigate the efficiency of 2-methoxyestradiol (2-ME) as radiosensitizing agent for the treatment of lung cancer cells. METHODS:Cell line A549 and GLC-82 originated from human non-small cell lung cancer were cultured in vitro. Study group (2-ME in different concentrations) and control group without 2-ME were set up. Cell proliferation was measured by MTT assay that lung cancer cells were treated with 2-ME for 24 h,then the cells were exposed from 0 to 8Gy radiation,and the survival fraction was determined by clone forming test. Flow cytometry was used to measure the effects of 2-ME on cell cycle distribution. RESULTS:MTT assay showed minimum effective concentration value was 0.15625×10~(-6) mol/L in GLC-82 and 1.25×10~(-6) mol/L in A549 cells. Compared to control group,exposed GLC-82 cells or A549 cells to minimum effective concentration of 2-ME for 24 h before irradiation resulted in an enhancement of radiation. The protection enhancement factor was 1.98 and 2.06 in GLC-82 and A549 cells,respectively. Flow cytometry analysis of cell cycle progression demonstrated G_2/M phase arrest in both cells in a dose dependent manner. No obvious change of CDK2 activity in both GLC-82 cells and A549 cells was observed. CONCLUSION:2-ME enhances radiosensitivity by G_2/M phase arrest in the cell cycle.

ABSTRACT

Objective To clone the coding dense granules protein 8(GRA8) from Toxoplasma gondii RH isolate for the potential use in the development of DNA vaccination. Methods Amplifing gene fragment coding GRA8 from the genomic DNA of Toxoplasma gondii RH isolate by means of ploymerase chain reaction (PCR), the gene is inserted into cloning vector pUC-19 digesting with restrictive enzymes and linking react ions. The positive colon is screed on LB plates containing ampicilline and IPTG, Xgal identified by blue-white and restrictive enzyme digestion. The inserted GR A8 gene was recombined with pcDNA3.1(+) eukaryotic expression vector by digestion with restrictive enzyme and linking reactions. The positive coloe is screene d o n LB plates containing ampicillin and identified by restrictive enzymes and link ing reactions. Results The GRA8 gene with about 804 base is specifically amplified from genomic DNA of Toxoplasma GRA8/RH and pcDNA3.1(+)-GRA8 recombinant is successfull y constructed. The sequencing results showed that GRA8 gene of isolate RH and R H from genebank shares quite high homology. Conclusion The gene encoding GRA8 is amplified from genomic DNA of Toxoplasma gene isolate GRA8/RH and pcDNA3.1 (+)-GRA8 recombinant is successfully constructed.

ABSTRACT

In this study, immunotoxin (IT) was prepared by conjugating BAC5 and CT with SPDP. The effects of IT on NPC and its mechanisms were explored using double labeled with radioactive nuclides, immunography and electron microscope technique in vivo and in vitro. The specific concentration of BAC5 in the tumor area showed. The radioactivity rate of tumor/nontumor (T/NT) was up to 10.26. IT had cytotoxic effects both on the cultured CNE-2 cell line and tumor multicell spheroides. In vivo, the preliminary result indicated that IT also had a inhibitory action on the nude mice models bearing human NPC (Reported in another article). Under electron microscope, the necrosis and apoptosis of tumor cells were found. The membranes of most tumor cells were found intacted not or corrosined, some of them had the character of apoptosis, including reduce of tumor cells membrane villi, condensation of cytoplasm and pyknosis or cleavage of nuclear. There were many of apoptosis bodies, which were occasionally phagocytosed by tumor cells. The infiltration of immunocytoes in tumor tissue could be seen. The results indicated that BAC5 can specifically combine with NPC cells and BAC5-CT has the inhibitory effect on NPC in vitro and in vivo, mechanism of which may be related to the effects that ‘warhead' CT dissolute the membrane of tumor cells directly, or/and IT promote the infiltration of immunocytoes so as to induce the apoptosis of tumor cell.

ABSTRACT

AIM:To observe the antineoplastic effect of Naja Naja atra venom (NNAV) on S180 bearing mice and to study the possible antineoplastic mechanism. METHODS:We observed the effect of NNAV on tumor weight、plasma nitric oxide content、plasma endothelin content and spleen index in S180 bearing mice with different concentration and different period by means of injecting into abdomen. RESULTS:Treatment with NNAV solution of different concentration could markedly inhibit S180 growth (especially in the low concentration group and by long period) and rate of inhibiting ranged from 21 63% to 49.25%; the plasma nitric oxide content, the plasma endothelin content and NO/ET ratio in tumor bearing mice were obviously higher than those of the normal control group, while after treatment with NNAV solution, the plasma nitric oxide level, the plasma endothelin level and NO/ET ratio could be reduced markedly, and it was noticed that NO/ET ratio in the group with highest inhibiting rate was most close to that of the normal control group. The spleen index was obviously increased after treatment with NNAV solution.CONCLUSION:The antineoplastic effect of NNAV on S180 bearing mice is best in long period by means of injecting into abdomen with low concentration. The mechanism of the antineoplastic effect of NNAV may be related to lowering the plasma nitric oxide and endothelin level, regulating the NO/ET ratio and enhancing the immune response.

ABSTRACT

AIM: To investigate the changes of plasma glutathione peroxidase (GSH-PX) and catalase (CAT) activities in nude mice (NM) bearing human nasopharyngeal carcinoma (NPC) and observe the effect of naja naja atra venom (NNAV) on them. METHODS: Plasma GSH-PX and CAT activities in human NPC bearing NM treated ( i.p. ) by low, middle or high concentration NNAV solution (1 mg/L, 5 mg/L, 10 mg/L) were determined by colorimetry. RESULTS: Plasma CAT activity (16 450 U/L) in NM bearing tumor group decreased significantly in comparison with the control group (20 680 U/L)(P0.05). Treated by low, middle or high concentration NNAV solution, CAT activities of three NM bearing tumor groups (20 570 U/L, 23 090 U/L, 21 280 U/L ) were higher than that of the NM bearing tumor group without NNAV treatment (16 450 U/L) (P0.05). ABSTRACT

AIM:To investigate the correlation between cerebral glucose metabolism and paediatric epilepsy or seizure type and seizure frequency.METHODS:To observe the cerebral glucose metabolism in epileptogenic focus,18F-FDG positron emission computed tomography(PET) brain imaging tests were carried out in 86 cases of paediatric epilepsy diagnosed by EEG and MRI.RESULTS:Compared to control group,significantly statistical differences between children epilepsy group and control group in PET brain imaging were observed(P0.05) was observed.CONCLUSION:Cerebral glucose metabolism of paediatric epilepsy is abnormal.The abnormal PET with varieties of epilepsy is found in different brain district.There is positive correlation between the abnormal intensity of PET imaging and the severity or seizure frequency of epilepsy.

ABSTRACT

AIM: To investigate the angiogenesis in the process of sarcoma 180 (S_(180)) tumor transplantation and changes of regulator factors, and explore the possible mechanism. METHODS: The S_(180) transplanted tumor in the Km mouse was used to detect the tumor angiogenesis by immunohistochemical examination of FⅧ. The levels of VEGF (V) and endostatin (E) in serum and the homogenate of tumor tissue were measured by ELISA and EIA, and the correlation between tumor weight and microvessel count (MVC) and morphology in tumor was also analyzed by multiple ANNOVA method. RESULTS: MVC, the relative count of total vessels and relative total vessel area increased with the development of transplanted S_ 180 . VEGF level in tumor tissue were higher at the 10th and 15th day than the 5th day after tumor transplantation. Endostatin in the tumor tissue and serum both reached the highest level at the 15th day, V/E ratio did not changed in this process. Furthermore, MVC, average vessel area and relative total area had a significant correlation with tumor weight. CONCLUSION: MVC increases in the development of S_(180) transplantation tumor and is related with the tumor weight; the positive regulator of angiogenesis in the tumor tissue is up-regulated during tumor growth, and the regulators in the tumor tissue maintains a relative balance.

AIM: To investigate the anticancer effect of manumycin on abdominal metastatic breast cancer cell line-SK-BR-3 and its relationship with p38 MAPK . METHODS: The test of anticancer effect was performed by the method of MTT, apoptosis induced by manumycin and affected by SB203580, a specific p38 MAPK inhibitor, were examined by caspase-3 activity assay kit, and the protein expression was detected by immunoblotting assay. RESULTS: The inhibition rates at 24 h after treatment with manumycin of 6 ?mol/L, 18 ?mol/L, 54 ?mol/L were (7.4?3.9)%, (21.0?4.4)% and (64.7?4.1)%, respectively and showed dosage-effect relationship. ABSTRACT

AIM: To investigate the protective effect of anti-macrophage migration factor monoclonal antibody (anti-MIF MAb) on oleic-acid-induced acute lung injury (ALI) rats and its influence on the expression level of MIF and intercellular adhesion molecule-1(ICAM-1). METHODS: The rats were subjected to injection of oleic acid (oleic acid group) or saline solution (control group). One hours before administration of oleic acid, the rats were intraperitoneally injected with anti-MIF antibody (5 mg/kg) as the treatment group. After injecting oleic acid or saline for 4 hours, the PaO_2, lung permeability index (LPI), the number of macrophage and the level of soluble ICAM-1 (sICAM-1) in the bronchial alveolar lavage fluid (BALF) were measured. The expression level of MIF mRNA and ICAM-1 mRNA in the lung were detected by in situ hybridization, and the degree of macrophage infiltration and the expression of MIF were evaluated by double staining immunocytochemistry. ABSTRACT

Nasopharyngeal carcinoma (NPC) in BALB/c nude mice (NM) was re-produced by hypodermic implanting human CNE-2 cell to the right-side of neck-backposi-tion. The changes of plasma malondialdehyde (MDA) and cAMP/cGMP ratio in NM bea-ring human NPC were examined. Meanwhile, we observed that tumor's size, plasma MDAand cAMP/cGMP ratio in NM bearing human NPC were affected by polysacchoridopetide(PSP). Results indicated that in comparison with the control group, NM bearing humanNPC showed a marked increase in plasma MDA(18.25?1.37 nmol/ml),cAMP/cGMP ratio(4.55?0.35), a marked decrease in plasma, cAMP (71.83?8.51 pM/ml) and cGMP(15.77?0.65 pM/ml), both high or low-concentration PSP inhibited NPC growth.Rate of inhibition was between 59.58 to 95.53 percent. PSP decreased plasma MDA,elevated cAMP, cGMP's levels and recovered cAMP/cGMP ratio. Our results suggestedthat the elevated level of MDA in NM bearing human NPC was closely associated withthe presence of NPC. PSP had antitumor effect, probably by means of decreasing freeradicals and recovering cAMP/cGMP ratio.
